| I. Background | |
| 生长分化因子 15（GDF15），也称为巨噬细胞抑制性细胞因子 1（MIC-1），属于 TG F-β超家族蛋白。GDF15 是一种内分泌激素，主要参与细胞生长、分化和组织修复。GDF1 5 作为一种炎症标志物，在肿瘤、缺血性疾病、代谢紊乱和神经退行性过程的发病机制中 发挥作用。 GFRAL（GDNF-family receptor α-like），也被称为 GRAL，属于胶质细胞源性神经营 养因子家族α样受体。GFRAL 是一种仅在后脑中表达的跨膜蛋白，需要辅助蛋白 RET 的帮 助才能进行信号传导。当 GDF15 与 GFRAL 结合时，RET 将自磷酸化并导致下游信号通路 （如 PI3K-AKT 和 PLC-PKC）的激活，从而调节许多生理过程，包括影响食物摄入和导致 体重减轻。抑制 GDF15-GFRAL 信号通路则能够增强免疫系统对实体瘤的杀伤，并能够治 疗或预防癌症恶病质。 |
